    Crossover Behavior from Decoupled Criticality

    We study the thermodynamic phase transition of a spin Hamiltonian comprising two 3D magnetic sublattices. Each sublattice contains XY spins coupled by the usual bilinear exchange, while spins in different sublattices only interact via biquadratic exchange. This Hamiltonian is an effective model for XY magnets on certain frustrated lattices such as body centered tetragonal. By performing a cluster Monte Carlo simulation, we investigate the crossover from the 3D-XY fixed point (decoupled sublattices) and find a systematic flow toward a first-order transition without a separatrix or a new fixed point. This strongly suggests that the correct asymptotic behavior is a first-order transition.Comment: 10 pages, 3 figures; added reference

    New NTP analogs: the synthesis of 4′-thioUTP and 4′-thioCTP and their utility for SELEX

    The synthesis of the triphosphates of 4′-thiouridine and 4′-thiocytidine, 4′-thioUTP (7; thioUTP) and 4′-thioCTP (8; thioCTP), and their utility for SELEX (systematic evolution of ligands by exponential enrichment) is described. The new nucleoside triphosphate (NTP) analogs 7 and 8 were prepared from appropriately protected 4′-thiouridine and -cytidine derivatives using the one-pot method reported by J. Ludwig and F. Eckstein [(1989) J. Org. Chem., 54, 631–635]. Because SELEX requires both in vitro transcription and reverse transcription, we examined the ability of 7 and 8 for SELEX by focusing on the two steps. Incorporation of 7 and 8 by T7 RNA polymerase to give 4′-thioRNA (thioRNA) proceeded well and was superior to those of the two sets of frequently used modified NTP analogs for SELEX (2′-NH(2)dUTP and 2′-NH(2)dCTP; 2′-FdUTP and 2′-FdCTP), when an adequate leader sequence of DNA template was selected. We revealed that a leader sequence of about +15 of DNA template is important for the effective incorporation of modified NTP analogs by T7 RNA polymerase. In addition, reverse transcription of the resulting thioRNA into the complementary DNA in the presence of 2′-deoxynucleoside triphosphates (dNTPs) also proceeded smoothly and precisely. The stability of the thioRNA toward RNase A was 50 times greater than that of the corresponding natural RNA. With these successful results in hand, we attempted the selection of thioRNA aptamers to human α-thrombin using thioUTP and thioCTP, and found a thioRNA aptamer with high binding affinity (K(d) = 4.7 nM)

    Probabilistic tsunami hazard assessment based on the Gutenberg–Richter law in eastern Shikoku, Nankai subduction zone, Japan

    Earthquake and tsunami predictions comprise huge uncertainties, thus necessitating probabilistic assessments for the design of defense facilities and urban planning. In recent years, computer development has advanced probabilistic tsunami hazard assessments (PTHAs), where hazard curves show the exceedance probability of the maximum tsunami height. However, owing to the lack of historical and geological tsunami records, this method is generally insufficient for validating the estimated hazard curves. The eastern coast of Shikoku in the Nankai subduction zone, Japan, is suitable for validation because tsunami records from historical Nankai Trough earthquakes are available. This study evaluated PTHAs by comparing the tsunami hazard curves and exceedance frequencies of historical Nankai Trough tsunamis. We considered 3480 earthquake scenarios representing the rupture patterns of past Nankai earthquakes and calculated all tsunamis. The probability of earthquake occurrence was based on the Gutenberg–Richter law. We considered uncertainty in tsunami calculations with astronomical tide variations. The estimated tsunami hazard curves are consistent with the exceedance frequencies obtained from historical tsunamis. In addition, sensitivity tests indicate the significance of the earthquake slip heterogeneity and tsunami defense facilities in PTHAs. We also extended the PTHAs to tsunami inundation maps in high resolution and proposed an effective new method for reducing the tsunami computation load

    Symbiotic Growth of a Thermophilic Sulfide-Oxidizing Photoautotroph and an Elemental Sulfur-Disproportionating Chemolithoautotroph and Cooperative Dissimilatory Oxidation of Sulfide to Sulfate

    A thermophilic filamentous anoxygenic photosynthetic bacterium, Chloroflexus aggregans, is widely distributed in neutral to slightly alkaline hot springs. Sulfide has been suggested as an electron donor for autotrophic growth in microbial mats dominated with C. aggregans, but remarkable photoautotrophic growth of isolated C. aggregans has not been observed with sulfide as the sole electron source. From the idea that sulfide is oxidized to elemental sulfur by C. aggregans and the accumulation of elemental sulfur may have an inhibitory effect for the growth, the effects of an elemental sulfur-disproportionating bacterium that consumes elemental sulfur was examined on the autotrophic growth of C. aggregans, strain NA9-6, isolated from Nakabusa hot spring. A sulfur-disproportionating bacterium, Caldimicrobium thiodismutans strain TF1, also isolated from Nakabusa hot spring was co-cultured with C. aggregans. C. aggregans and C. thiodismutans were successfully co-cultured in a medium containing thiosulfate as the sole electron source and bicarbonate as the sole carbon source. Quantitative conversion of thiosulfate to sulfate and a small transient accumulation of sulfide was observed in the co-culture. Then the electron source of the established co-culture was changed from thiosulfate to sulfide, and the growth of C. aggregans and C. thiodismutans was successfully observed with sulfide as the sole electron donor for the autotrophic growth of the co-culture. During the cultivation in the light, simultaneous consumption and accumulation of sulfide and sulfate, respectively, were observed, accompanied with the increase of cellular DNAs of both species. C. thiodismutans likely works as an elemental sulfur scavenger for C. aggregans, and C. aggregans seems to work as a sulfide scavenger for C. thiodismutans. These results suggest that C. aggregans grows autotrophically with sulfide as the electron donor in the co-culture with C. thiodismutans, and the consumption of elemental sulfur by C. thiodismutans enabled the continuous growth of the C. aggregans in the symbiotic system.
